Circular RNAs (circRNAs) are a sort of long, non-coding RNA molecules with a covalently closed continuous ring structure without 5'-3' polarity and poly-A tail. The modulative role of circRNAs in malignant diseases has been elucidated by many studies in recent years bioinformatics and high-throughput sequencing technologies. Generally, circRNA affects the proliferative, invasive, and migrative capacity of malignant cells various mechanisms, exhibiting great potential as novel biomarkers in the diagnoses or treatments of malignancies. Meanwhile, autophagy preserves cellular homeostasis, serving as a vital molecular process in tumor progression. Mounting studies have demonstrated that autophagy can not only contribute to cancer cell survival but can also induce autophagic cell death in specific conditions. A growing number of research studies have indicated that there existed abundant associations between circRNAs and autophagy. Herein, we systemically reviewed and discussed recent studies on this topic in different malignancies and concluded that the circRNA-autophagy axis played crucial roles in the proliferation, metastasis, invasion, and drug or radiation resistance of different tumor cells.
环状RNA(circRNAs)是一类长链非编码RNA分子,具有共价闭合的连续环状结构,没有5'-3'极性和多聚A尾。近年来,通过生物信息学和高通量测序技术,许多研究已经阐明了circRNAs在恶性疾病中的调节作用。一般来说,circRNA通过多种机制影响恶性细胞的增殖、侵袭和迁移能力,在恶性肿瘤的诊断或治疗中作为新型生物标志物具有巨大潜力。同时,自噬维持细胞内稳态,是肿瘤进展中的一个重要分子过程。越来越多的研究表明,自噬不仅有助于癌细胞存活,在特定条件下还能诱导自噬性细胞死亡。越来越多的研究表明,circRNAs与自噬之间存在丰富的关联。在此,我们系统地回顾和讨论了不同恶性肿瘤中关于该主题的最新研究,并得出结论:circRNA-自噬轴在不同肿瘤细胞的增殖、转移、侵袭以及药物或辐射抗性中发挥着关键作用。
