The effects of dietary supplementation with citrus flavonoid extract (CFE) on milk performance, rumen fermentation, rumen microbiome, rumen metabolome, and serum antioxidant indexes were evaluated. Eight multiparous lactating cows were allocated to a replicated 4 × 4 Latin square with 25-d periods consisting of 20 d of adaptation and 5 d of sampling. Experimental treatments included a control diet (CON) and CON supplemented with 50 g d (CFE50), 100 g d (CFE100), and 150 g d (CFE150). Feeding CFE to dairy cows increased milk production and milk lactose. Milk somatic cell count linearly reduced with increasing CFE amount. Supplementing CFE linearly increased the ruminal concentrations of total volatile fatty acids, acetate, propionate, butyrate, and microbial crude protein. Ruminal lipopolysaccharide linearly decreased with increasing CFE amount. Compared with CON, CFE150 cows exhibited a greater abundance of Firmicutes and a low abundance of Bacteroidetes. Cellulolytic bacteria (genera , , and ) and carbohydrate metabolism were enriched in the CFE150 cows. For archaea and viruses, major methanogens (genera and ) and phylum Uroviricota were inhibited in the CFE150 cows. Compared with CON, the ruminal concentrations of tyrosine, proline, pyruvate, glucose, and glucose-6-phosphate were higher in the CFE150 cows. The metabolites of citrus flavonoids, such as hippuric acid, hesperetin, and naringenin, were increased in the CFE150 cows. Supplementing CFE significantly improved the antioxidant capacity of the dairy cows. This study highlighted that dietary supplementation with CFE led to significant changes in the rumen microbial composition and metabolites, and consequently resulted in an improved lactational performance of dairy cows.
本研究旨在评估日粮中添加柑橘类黄酮提取物(CFE)对奶牛产奶性能、瘤胃发酵、瘤胃微生物区系、瘤胃代谢组和血清抗氧化指标的影响。选择 8 头经产泌乳奶牛,采用 4×4 拉丁方设计,每个处理 4 个重复,每个重复 1 头奶牛,每个周期 25d,其中预试期 20d,正试期 5d。试验设 1 个对照组(CON)和 3 个 CFE 处理组(CFE50、CFE100 和 CFE150),CFE 添加量分别为 50、100 和 150 g/d。结果表明,与 CON 组相比,CFE 组奶牛产奶量和乳中乳糖含量显著增加,乳体细胞数线性降低。CFE 组奶牛瘤胃总挥发性脂肪酸、乙酸、丙酸、丁酸和微生物蛋白浓度线性增加,瘤胃内毒素(LPS)浓度线性降低。与 CON 组相比,CFE150 组奶牛瘤胃厚壁菌门(Firmicutes)相对丰度增加,拟杆菌门(Bacteroidetes)相对丰度降低。CFE150 组奶牛中纤维素分解菌(属、和 )和碳水化合物代谢相关菌属丰度增加。在古菌和病毒方面,CFE150 组奶牛中主要产甲烷菌(属 和 )和 Uroviricota 门丰度降低。与 CON 组相比,CFE150 组奶牛瘤胃液中酪氨酸、脯氨酸、丙酮酸、葡萄糖和葡萄糖-6-磷酸浓度升高。CFE150 组奶牛中柑橘类黄酮的代谢产物如马尿酸、橙皮苷和柚皮苷等增加。CFE 添加显著提高了奶牛的抗氧化能力。综上,日粮添加 CFE 可显著改变奶牛瘤胃微生物组成和代谢物,从而提高奶牛泌乳性能。
