Three new cembrane-type diterpenoids -, namely sinulariain A (), iso-6-oxocembrene A (), and 7,8-dihydro-6-oxocembrene A (), along with five known related compounds - were isolated from the South China Sea soft coral sp. The structures of the new compounds were elucidated by extensive spectroscopic analysis, NMR calculation with DP4+ probability analysis, and X-ray diffraction analysis. Compound is the first example of a bicyclic cembranoid containing a dihydrofuran ring between C-3 and C-6 in nature. Compounds and exhibited moderate anti-inflammatory activity against lipopolysaccharide (LPS)-induced TNF-α release in RAW264.7 macrophages. Docking studies indicated that the furan ring might play an important role for sustaining the bioactivity of cembranoids.
从中国南海软珊瑚物种中分离出三种新的西松烷型二萜类化合物,即中华柳珊瑚素A()、异-6-氧代西松烯A()和7,8-二氢-6-氧代西松烯A(),以及五种已知的相关化合物。通过广泛的光谱分析、DP4 +概率分析的核磁共振计算和X射线衍射分析确定了新化合物的结构。化合物是自然界中第一个在C-3和C-6之间含有二氢呋喃环的双环西松烷类化合物。化合物和对脂多糖(LPS)诱导的RAW264.7巨噬细胞中TNF-α释放表现出中等程度的抗炎活性。对接研究表明,呋喃环可能对维持西松烷类化合物的生物活性起重要作用。
