The present study aimed to investigate the therapeutic effect of fullerene C nanoparticle against heart tissue damage caused by 7,12-dimethylbenz [a] anthracene (DMBA) in female rats. Female Wistar albino rats, 8 weeks old (n = 60) weighing around (150 ± 10 g) were used for the study. These rats were divided into 4 groups and each group included 15 rats. Groups: (i) Control Group: Fed with standard diet; (ii) C Group: C (1.7 mg/kg bw, oral gavage); (iii) DMBA Group: DMBA (45 mg/kg bw, oral gavage); (iv) C and DMBA Group: C (1.7 mg/kg bw, oral gavage) and DMBA (45 mg/kg bw, oral gavage) group. Malondialdehyde (MDA) analysis, catalase activity (CAT), and glutathione (GSH) in heart tissue were determined by spectrophotometer. In addition, heart tissue DNA damage was investigated. Caspase-3, p53, HO-1, COX-2, and TNF-α protein expression levels in heart tissue were determined by western blotting. As a result, Caspase-3, p53, HO-1 protein expression, GSH levels and CAT activity increased, COX-2, TNF-α protein expression, and MDA levels were significantly decreased in the C + DMBA group compared to the DMBA group. Therefore, the fullerene C nanoparticle may be a promising and effective therapy for the treatment of heart diseases associated with inflammation.
本研究旨在探讨富勒烯C纳米颗粒对7,12-二甲基苯并[a]蒽(DMBA)所致雌性大鼠心脏组织损伤的治疗作用。选用8周龄(n = 60)、体重约(150±10 g)的雌性Wistar白化大鼠进行研究。将这些大鼠分为4组,每组15只。分组如下:(i)对照组:喂以标准饮食;(ii)C组:C(1.7 mg/kg体重,经口灌胃);(iii)DMBA组:DMBA(45 mg/kg体重,经口灌胃);(iv)C和DMBA组:C(1.7 mg/kg体重,经口灌胃)和DMBA(45 mg/kg体重,经口灌胃)组。用分光光度计测定心脏组织中的丙二醛(MDA)含量、过氧化氢酶活性(CAT)和谷胱甘肽(GSH)水平。此外,还研究了心脏组织的DNA损伤情况。通过蛋白质免疫印迹法测定心脏组织中半胱天冬酶-3、p53、血红素加氧酶-1(HO-1)、环氧化酶-2(COX-2)和肿瘤坏死因子-α(TNF-α)的蛋白表达水平。结果显示,与DMBA组相比,C + DMBA组的半胱天冬酶-3、p53、HO-1蛋白表达、GSH水平和CAT活性升高,COX-2、TNF-α蛋白表达及MDA水平显著降低。因此,富勒烯C纳米颗粒可能是治疗与炎症相关心脏病的一种有前景且有效的疗法。
