Abstract

Sagittatoside B is one of the principal diglucosides in Herba Epimedii. In this work, an ultra-performance liquid chromatography/quadrupole-time-of-flight mass spectrometry (UPLC/Q-TOF-MS) was applied to the rapid analysis of sagittatoside B metabolites in rats after oral administration. A total number of 17 metabolites were detected or tentatively identified from rat plasma, bile, urine and feces. The major metabolic pathways of sagittatoside B in rats were hydrolysis, hydrogenation, hydroxylation, dehydrogenation, demethylation, decarbonylation and conjugation with glucuronic acid and different sugars. This work revealed the metabolism of sagittatoside B , and reported the characteristic metabolic reactions of sagittatoside B for the first time. This provided the basis for the further research and development of sagittatoside B, and also provided reference for the metabolism of active flavonoid compounds.

箭叶淫羊藿苷 B 是淫羊藿中的主要双糖苷之一。在这项工作中,采用超高效液相色谱/四极杆飞行时间质谱(UPLC/Q-TOF-MS)快速分析了口服给药后大鼠体内箭叶淫羊藿苷 B 的代谢产物。从大鼠血浆、胆汁、尿液和粪便中共检测或推测出 17 种代谢产物。箭叶淫羊藿苷 B 在大鼠体内的主要代谢途径为水解、氢化、羟基化、脱氢、去甲基化、脱羰基和与葡萄糖醛酸及不同糖结合。本研究揭示了箭叶淫羊藿苷 B 的代谢途径,并首次报道了箭叶淫羊藿苷 B 的特征代谢反应。这为箭叶淫羊藿苷 B 的进一步研究和开发提供了依据,也为活性黄酮类化合物的代谢提供了参考。
