A membrane-diffusion cell for oxygen uptake measurements in an open system is described. Polarographic oxygen measurements with a Clark electrode are combined with simultaneous pH and absorbance or fluorescence determinations. Examples of its use are given and discussed, including experiments on mitochondrial respiration. Advantages of this cell are analyzed and comparisons are made with other open-system respirographs.
