Abstract

A novel and efficient palladium-catalyzed annulation of anilines with bromoalkynes for the synthesis of 2-phenylindoles has been described. This approach features excellent regio- and stereoselectivities and good functional group tolerance. Preliminary mechanistic studies indicate that anilines undergo -nucleophilic addition to bromoalkynes to generate ()--(2-bromo-1-phenylvinyl) anilines, followed by sequential C-H functionalization to deliver different substituted 2-phenylindoles. This method provides potential applications for the construction of various biologically active compounds.

一种新型高效的钯催化苯胺与溴代炔烃的环化反应,用于合成 2-苯基吲哚。该方法具有优异的区域和立体选择性以及良好的官能团容忍性。初步的机理研究表明,苯胺与溴代炔烃发生 -亲核加成反应生成 ()--(2-溴-1-苯基乙烯基)苯胺,然后进行连续的 C-H 官能团化,得到不同取代的 2-苯基吲哚。该方法为构建各种生物活性化合物提供了潜在的应用。
