Abstract

OBJECTIVE

To investigate the relationships of leukemia inhibitory factor receptor (LIFR) with cervical cancer invasion and metastasis.

METHODS

From January 2021 to December 2022, 45 patients treated for cervical cancer and lung metastases were identified. Western blotting was used to determine the expression of Hippo-YAP signaling pathway-related proteins. Meanwhile, 40 healthy Sprague-Dawley nude mice were used and evenly randomized into two groups, which were injected with LIFR-overexpressing (study group) or normal cervical cancer cells (control group). The lung tissue of nude mice was removed for hematoxylin-eosin staining, and the number of lung cell metastases in nude mice was counted.

RESULTS

The highest LIFR mRNA expression was found in paracancerous tissue, followed by cervix cancer tissue and metastatic lesions. The study group exhibited higher LIFR, P-YAP, and P-TAZ protein expression and lower YAP and TAZ protein expression than the control group. The study group had a lower number of lung metastases than the control group.

CONCLUSION

Decreased expression of LIFR and decreased phosphorylation of Hippo-YAP signaling pathway-related proteins might be the underlying mechanisms that promote lung metastasis of cervical cancer.

目的

探讨白血病抑制因子受体(LIFR)与宫颈癌侵袭转移的关系。

方法

选取 2021 年 1 月至 2022 年 12 月期间收治的宫颈癌肺转移患者 45 例,采用 Western blot 法测定 Hippo-YAP 信号通路相关蛋白的表达。同时选取 40 只健康的 Sprague-Dawley 裸鼠,采用随机数字表法分为两组,分别注射过表达 LIFR(研究组)或正常宫颈癌细胞(对照组),取出裸鼠的肺组织行苏木精-伊红(HE)染色,计数裸鼠肺细胞转移数。

结果

LIFRmRNA 表达水平最高的为癌旁组织,其次为宫颈癌组织和转移灶;研究组 LIFR、P-YAP 和 P-TAZ 蛋白表达水平高于对照组,YAP 和 TAZ 蛋白表达水平低于对照组;研究组肺转移数少于对照组。

结论

LIFR 表达降低和 Hippo-YAP 信号通路相关蛋白磷酸化降低可能是促进宫颈癌肺转移的潜在机制。
