To investigate the effects of (SQWF) Formula on the NLRP3/GSDMD signaling pathway in pulmonary deficiency syndrome rats with chronic obstructive pulmonary diseases (COPD).
Rat models of COPD and lung deficiency syndrome induced by exposure to cigarette smoke and lipopolysaccharide (LPS) injection were randomized (=8) for treatment with low-, medium- and high-dose SQWF or (YPF). The changes in body weight, grip strength, lung function, pulmonary pathology, peripheral blood inflammatory cell counts, and levels of inflammatory factors in the bronchoalveolar lavage fluid (BALF) were examined. The expressions of proteins associated with the NLRP3/GSDMD signaling pathway in the lung tissues were determined with RT-qPCR and immunohistochemistry.
The rat models of COPD and lung deficiency syndrome showed significantly reduced body weight, grip strength and lung function (<0.01) with severe lung pathologies, increased levels of WBC, NEU% and MON% (<0.01), decreased LYM% (<0.01), and increased levels of TNF-α, IL-6, IL-1β and IL-18 in the BALF (<0.01); the mRNA and protein expression levels of NLRP3, ASC, GSDMD and IL-1β all increased significantly in the lung tissue (<0.01). Treatment with SQWF and YPF obviously increased body weight and improved grip strength, pulmonary function and lung pathology of the COPD rats (<0.05). The treatments obviously improved the changes in peripheral blood inflammatory cell counts and inflammatory factors in the BALF of the rat models (<0.01) and lowered the expressions of NLRP3, ASC, GSDMD and IL-1β in the lung tissues (<0.05) without significantly affecting the mRNA levels of caspase-1 and IL-18 (>0.05), and the effects of SQWF were dose-dependent.
SQWF Formula relieves inflammatory response and injury in the lung tissue of COPD rats with pulmonary qi deficiency syndrome possibly by inhibiting pyroptosis through regulating the NLRP3/GSDMD pathway.
探讨四君子加味方(SQWF)对慢性阻塞性肺疾病(COPD)肺气虚证大鼠NLRP3/GSDMD信号通路的影响。
将通过暴露于香烟烟雾和注射脂多糖(LPS)诱导的COPD和肺气虚证大鼠模型随机分为8组,分别用低、中、高剂量的SQWF或玉屏风方(YPF)进行治疗。检测体重、握力、肺功能、肺病理学、外周血炎症细胞计数以及支气管肺泡灌洗液(BALF)中炎症因子水平的变化。用RT-qPCR和免疫组织化学法测定肺组织中与NLRP3/GSDMD信号通路相关蛋白的表达。
COPD和肺气虚证大鼠模型体重、握力和肺功能显著降低(P<0.01),伴有严重的肺病理学改变,白细胞、中性粒细胞百分比和单核细胞百分比水平升高(P<0.01),淋巴细胞百分比降低(P<0.01),BALF中肿瘤坏死因子-α、白细胞介素-6、白细胞介素-1β和白细胞介素-18水平升高(P<0.01);肺组织中NLRP3、凋亡相关斑点样蛋白(ASC)、Gasdermin D(GSDMD)和白细胞介素-1β的mRNA和蛋白表达水平均显著升高(P<0.01)。SQWF和YPF治疗明显增加了COPD大鼠的体重,改善了握力、肺功能和肺病理学(P<0.05)。治疗明显改善了大鼠模型外周血炎症细胞计数和BALF中炎症因子的变化(P<0.01),降低了肺组织中NLRP3、ASC、GSDMD和白细胞介素-1β的表达(P<0.05),且对半胱天冬酶-1和白细胞介素-18的mRNA水平无明显影响(P>0.05),SQWF的作用呈剂量依赖性。
四君子加味方可能通过调节NLRP3/GSDMD途径抑制细胞焦亡,从而减轻COPD肺气虚证大鼠肺组织的炎症反应和损伤。
