Cysteine (Cys) is one of the most basic mercaptans in the human body. As an important endogenous small molecule mercaptan, Cys plays a vital role in various physiological processes and can participate in maintaining redox balance to ensure homeostasis. Abnormal Cys levels can lead to a variety of diseases. However, the detection of cysteine may be interfered with by other small molecule biothiols. Therefore, the design of fluorescent probes based on the structural characteristics and reactivity of cysteine has become the focus of current research. In this paper, a fluorescent probe (3-(2-benzo[][1,2,3]triazol-2-yl)-2-oxo-2-benzo[]chromen-8-yl acrylate, BTAB) for Cys detection was synthesized with acrylic ester as the reaction site. Under the conditions of gradual optimization, BTAB can achieve selectivity and anti-interference ability for Cys detection. The linear range of Cys was 0.3-10 μM, and the detection limit was 0.154 μM. Finally, this probe was applied to detect the Cys content in bovine serum samples with satisfactory results.
半胱氨酸(Cys)是人体内最基本的巯基化合物之一。作为一种重要的内源性小分子巯基化合物,Cys 在各种生理过程中发挥着重要作用,能够参与维持氧化还原平衡以确保内环境稳定。Cys 水平的异常可导致多种疾病。然而,Cys 的检测可能会受到其他小分子生物硫醇的干扰。因此,基于 Cys 的结构特征和反应性设计荧光探针已成为当前研究的重点。在本文中,我们合成了一种以丙烯酯为反应位点的用于检测 Cys 的荧光探针(3-(2-苯并[][1,2,3]三唑-2-基)-2-氧代-2-苯并[]色烯-8-基丙烯酸酯,BTAB)。在逐步优化条件下,BTAB 能够实现对 Cys 检测的选择性和抗干扰能力。Cys 的线性范围为 0.3-10 μM,检测限为 0.154 μM。最后,该探针成功应用于牛血清样品中 Cys 含量的检测,结果令人满意。
