This study aimed to explore the effect and mechanism of Yunkang oral liquid (YK) on polycystic ovary syndrome (PCOS). PCOS model rats were prepared by injecting exogenous androgen dehydroepiandrosterone, and YK was administered simultaneously for 28 days during modeling. The morphology of ovaries and uterus was observed using H&E staining, and serum levels of testosterone (T), luteinizing hormone (LH), and follicle-stimulating hormone (FSH) were determined by radioimmunoassay. Additionally, serum lipids (TG, HDL-c), blood glucose (GLU), and aminotransferase (AST, ALT) levels were detected. The expression of androgen receptor (AR) protein was determined by Western blotting. YK treatment resulted in reduced serum levels of T, LH and FSH, ameliorated ovarian polycystic-like pathological changes and uterine morphology in PCOS rats, and decreased serum TG, GLU, AST and ALT levels, elevated serum HDL-c levels, and improved abnormalities of glycolipid metabolism accompanying PCOS. Moreover, YK decreased the expression of ovarian AR in PCOS rats. This study indicates that YK may protect the ovaries by inhibiting the expression of AR, which could be a potential treatment for PCOS.
本研究旨在探讨运康口服液（YK）对多囊卵巢综合征（PCOS）的作用及其机制。通过注射外源性雄激素脱氢表雄酮（DHEA）制备 PCOS 模型大鼠，同时在建模期间给予 YK 治疗 28 天。采用 H&E 染色观察卵巢和子宫的形态，放射免疫法测定血清睾酮（T）、黄体生成素（LH）和卵泡刺激素（FSH）水平。此外，检测血清脂质（TG、HDL-c）、血糖（GLU）和氨基转移酶（AST、ALT）水平。采用 Western blot 法检测雄激素受体（AR）蛋白的表达。YK 治疗可降低 PCOS 大鼠血清 T、LH 和 FSH 水平，改善卵巢多囊样病理变化和子宫形态，降低血清 TG、GLU、AST 和 ALT 水平，升高血清 HDL-c 水平，改善 PCOS 伴随的糖脂代谢异常。此外，YK 还可降低 PCOS 大鼠卵巢 AR 的表达。本研究表明，YK 可能通过抑制 AR 的表达来保护卵巢，这可能是 PCOS 的一种潜在治疗方法。
