To investigate the effect of Chinese medicine He's Yangchao recipe on premature ovarian insufficiency (POI) and its relationship with mitochondrial function of ovarian granulose cells in an animal model.
Thirty-six female C57BL/6J mice were randomly divided into blank control group, model group, low-, medium- and high-dose He's Yangchao recipe treatment group and coenzyme Q10 (Q10) treatment group (positive control). The POI model was induced by a single intraperitoneal injection of cyclophosphamide (90 mg/kg). The animals were sacrificed after 21 days. Primary granulose cells were obtained from POI mice and treated with He's Yangchao recipe, ERβ inhibitor PHTPP, and He's Yangchao recipe+PHTPP for 24 h, respectively. Ovarian histopathological changes were observed by hematoxylin-eosin (HE) staining, ATP levels were detected by luciferase assay, mtDNA copy numbers were detected by quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR), mitochondrial structure changes were observed by transmission electron microscopy, protein and mRNA expression levels of estrogen receptor β (ERβ), peroxisome proliferator-activated receptor γ coactivator 1α (PGC1α), mitochondrial transcription factor A (TFAM), and superoxide dismutase 2 (SOD2) were detected by Western blotting and qRT-PCR.
The ovarian tissue in model group exhibited few secondary and tertiary follicles, whereas the He's Yangchao recipe groups and Q10 group had abundant secondary and tertiary follicles. Compared with the blank control group, ATP and mtDNA levels in model group decreased (<0.01), mitochondrial crista disappeared or abnormal vacuolated structure increased; the protein and mRNA levels of ERβ, PGC1α, TFAM, and SOD2 decreased (all <0.01). ATP production increased in granulose cells of high-dose He's Yangchao recipe group and Q10 group; mtDNA copy numbers increased (<0.05 or <0.01); abnormal mitochondrial structure was reduced; the protein and mRNA expressions of ERβ, PGC1α, TFAM, and SOD2 increased (<0.05 or <0.01). Compared with the PHTPP intervention group, the proportion of normal mitochondrial structure in the granulose cells of He's Yangchao recipe + PHTPP group was higher; ATP content increased (<0.05 or <0.01); mtDNA copy numbers increased (<0.05 or <0.01); the protein and mRNA expression of ERβ, PGC1α, TFAM and SOD2 increased (<0.05 or <0.01).
He's Yangchao recipe can regulate mitochondrial biogenesis through ERβ/PGC1α/TFAM pathway to improve ovarian function in POI mice.

36 只雌性 C57BL/6J 小鼠随机分为空白对照组、模型组、和营振阳方低、中、高剂量组和辅酶 Q10(Q10)阳性对照组。采用单次腹腔注射环磷酰胺(90mg/kg)建立 POI 模型。21 天后处死动物。分离 POI 小鼠原代颗粒细胞,分别用和营振阳方、雌激素受体β(ERβ)抑制剂 PHTPP 和和营振阳方+PHTPP 处理 24h。采用苏木精-伊红(HE)染色观察卵巢组织病理变化,采用荧光素酶法检测三磷酸腺苷(ATP)水平,采用实时荧光定量聚合酶链反应(qRT-PCR)检测线粒体 DNA(mtDNA)拷贝数,采用透射电镜观察线粒体结构变化,采用 Western blot 和 qRT-PCR 检测 ERβ、过氧化物酶体增殖物激活受体γ共激活因子 1α(PGC1α)、线粒体转录因子 A(TFAM)和超氧化物歧化酶 2(SOD2)的蛋白和 mRNA 表达水平。
模型组卵巢组织中次级和三级卵泡较少,而和营振阳方组和 Q10 组次级和三级卵泡丰富。与空白对照组相比,模型组 ATP 和 mtDNA 水平降低(均<0.01),线粒体嵴消失或异常空泡状结构增加;ERβ、PGC1α、TFAM 和 SOD2 的蛋白和 mRNA 水平降低(均<0.01)。与模型组相比,高剂量和营振阳方组和 Q10 组颗粒细胞中 ATP 生成增加;mtDNA 拷贝数增加(均<0.05 或<0.01);异常线粒体结构减少;ERβ、PGC1α、TFAM 和 SOD2 的蛋白和 mRNA 表达增加(均<0.05 或<0.01)。与 PHTPP 干预组相比,和营振阳方+PHTPP 组颗粒细胞中正常线粒体结构的比例更高;ATP 含量增加(均<0.05 或<0.01);mtDNA 拷贝数增加(均<0.05 或<0.01);ERβ、PGC1α、TFAM 和 SOD2 的蛋白和 mRNA 表达增加(均<0.05 或<0.01)。
和营振阳方可通过 ERβ/PGC1α/TFAM 通路调节线粒体生物发生,改善 POI 模型小鼠的卵巢功能。
