The main aim of this study was to formulate and optimize sustained release mini-tablets of guaifenesin. Guaifenesin granules were successfully prepared using different blend ratios of carnauba wax to drug by melt granulation method. The properties of granules were further modified by combining them with ethyl cellulose. The obtained granules were then mixed and compressed into mini-tablets using a tablet press machine. The resulting mini-tablets were characterized in terms of weight, thickness, hardness, drug content and drug release. Mini-tablets with 1:6 carnauba wax to drug ratio showed superior physicochemical characteristics, releasing about 100.03% of guaifenesin over 8 h. Ethyl cellulose offers a great potential to accurately control drug release from mini-tablets. The prepared mini-tablets seem to be a very promising alternative to guaifenesin conventional formulations and can be used in adults and elderly people.
本研究的主要目的是制备并优化愈创甘油醚的缓控释微型片剂。采用熔融制粒法,成功制备了不同比例的巴西棕榈蜡与药物的愈创甘油醚颗粒。通过与乙基纤维素结合,进一步对颗粒的性质进行了修饰。然后,使用压片机将得到的颗粒混合并压制成微型片剂。所得微型片剂的重量、厚度、硬度、药物含量和药物释放度等方面进行了特征描述。当巴西棕桐蜡与药物的比例为 1:6 时,微型片剂表现出优越的理化特性,在 8 小时内释放了约 100.03%的愈创甘油醚。乙基纤维素为微型片剂的药物释放提供了一种非常有前途的精确控制方法。制备的微型片剂似乎是愈创甘油醚常规制剂的一种很有前途的替代品,可用于成年人和老年人。
