Abstract

BACKGROUND

Colibacillosis is an important epidemiological and economic issue in poultry farming and breeding. A common problem with avian pathogenic Escherichia coli strains (APEC) that cause this disease is a lack of an uniform identification system, resulting from a variety of serotypes, phylogenetic groups, sequence types and combinations of virulence factors. There are no clearly defined features that can be associated with pathogenicity. Therefore, without precise identification of pathogenic strains and differentiation from commensal strains, there is no possibility of appropriate selection of targeted therapy. The widespread use of whole genome sequencing (WGS) in recent years creates new possibilities in diagnostics. Therefore, the purpose of this study was to select features defining the APEC pathotype, based on next generation sequencing (NGS), and design a diagnostic test based on selected factors.

RESULTS

A PCR diagnostic test is proposed. Three predictors of virulence were chosen according to in silico analysis: two virulence genes: iroC and hlyF, as well as one molecular marker of O78 serotype (wzx-O-antigen flippase of the O78 serotype). A choice of markers was supported by a chicken embryo model.

CONCLUSIONS

Whole genome sequencing of E. coli genomes allowed for the development of a rapid diagnostic method identifying pathogenic strains for poultry: APEC. The developed test can support field observations connected with the strain isolation source and clinical symptoms of the disease.

背景

大肠杆菌病是家禽养殖和育种中的一个重要流行病学和经济问题。引起这种疾病的禽致病性大肠杆菌菌株(APEC)的一个常见问题是缺乏统一的鉴定系统,这是由于多种血清型、系统发育群、序列类型和毒力因子组合造成的。没有明确界定的可与致病性相关联的特征。因此,在没有精确鉴定致病菌株并将其与共生菌株区分开来的情况下,就不可能进行针对性治疗的恰当选择。近年来全基因组测序(WGS)的广泛应用为诊断创造了新的可能性。因此,本研究的目的是基于下一代测序(NGS)选择定义APEC致病型的特征,并根据所选因素设计一种诊断测试。

结果

结论
