It was shown that when the filtration capacity of the kidneys was lowered even insignificantly (the creatinine clearance 50-80 ml/min), the maximum blood levels of ampicillin increased 2 times and its retention time was prolonged. At the terminal stage of chronic renal insufficiency the half-life of the drug and the area under the pharmacokinetic curve markedly increased and the plasma clearance decreased. Hemodialysis and hemosorption significantly lowered the ampicillin half-life. The ampicillin coefficient in hemosorption was 2 times higher than that in hemodialysis.
结果表明,当肾脏滤过能力即使轻微降低(肌酐清除率为50 - 80毫升/分钟)时,氨苄西林的最高血药浓度增加2倍,其滞留时间延长。在慢性肾功能不全终末期,药物的半衰期和药代动力学曲线下面积显著增加,血浆清除率降低。血液透析和血液吸附可显著降低氨苄西林的半衰期。血液吸附中氨苄西林的系数比血液透析高2倍。
