Abstract

In this study, a SDU strain capable of producing poly-γ-glutamate (γ-PGA) was newly identified from the rhizosphere soil of Baimiao taro. The strain is a glutamate-independent strain and can produce polyglutamic acid in a culture medium completely free of glutamate. The hydrolyzed product of the polyglutamic acid produced is D-glutamic acid. The molecular weight of γ-PGA, estimated via the Mark-Houwink equation, was 1390 kDa. Furthermore, the molecular weight measured by Waters gel permeation chromatography with multi-angle laser light scattering (GPC-MALLS) was 1167 kDa. The production of γ-PGA and its antioxidant and tyrosine inhibition properties were investigated. The γ-PGA production reached 23.1 g/L, and the productivity was 0.77 g L h. Specifically, γ-PGA exhibited superoxide anion (·O) radical scavenging activity and tyrosinase inhibitory activity. This study introduces a promising strain and a highly efficient application method for γ-PGA, which can be broadly utilized in the pharmaceutical, food, and cosmetic industries.

在本研究中,从白庙芋头的根际土壤中新鉴定出一株能够产生聚γ-谷氨酸(γ-PGA)的菌株。该菌株是一株不依赖谷氨酸的菌株,能够在完全不含谷氨酸的培养基中产生聚谷氨酸。所产生的聚谷氨酸的水解产物为D-谷氨酸。通过Mark-Houwink方程估算,γ-PGA的分子量为1390 kDa。此外,采用多角度激光光散射沃特世凝胶渗透色谱法(GPC-MALLS)测得的分子量为1167 kDa。研究了γ-PGA的产生及其抗氧化和酪氨酸抑制特性。γ-PGA的产量达到23.1 g/L,生产率为0.77 g L h。具体而言,γ-PGA表现出超氧阴离子(·O)自由基清除活性和酪氨酸酶抑制活性。本研究介绍了一种有前景的γ-PGA菌株及其高效应用方法,其可广泛应用于制药、食品和化妆品行业。
