To investigate the protective effects of five alkylresorcinol homologs on hydrogen peroxide(H_2O_2)-induced oxidative damage in human colorectal adenocarcinoma cells(Caco-2).
The H_2O_2-induced oxidative damage of Caco-2 cells was used as a model, and the survival rate of Caco-2 cells pretreated with different concentrations(0-80 μmol/L)of 5-n-alkylresorcinols was determined to determine the optimal concentration range. The protective effects of 5-n-alkylresorcinols on oxidative-damaged cells was evaluated by measuring the levels of reactive oxygen species(ROS), malondialdehyde(MDA), glutathione peroxidase(GSH-Px), superoxide dismutase(SOD) and catalase(CAT) activities.
5-n-alkylresorcinols had no obvious toxic effect on Caco-2 cells in the concentration range of 5-80 μmol/L. The activities of GSH-Px, SOD and CAT oxidase in cell culture supernatant and cells were significantly lower than those in the control group after 4 hours of 600 μmol/L H_2O_(2 )treatment(P<0.05). The addition of 5 alkylresorcinol homologues at different concentrations alleviated the damage of H_2O_2 to different degrees, and the ROS level and MDA content decreased compared with the modularity, and the activity of GSH-Px, SOD and CAT was restored to a certain extent.
5-n-alkylresorcinols can protect Caco-2 cells from oxidative damage to H_2O_2 by activating intracellular antioxidant enzyme activity and inhibiting lipid peroxidation levels.
研究5种烷基间苯二酚同系物对过氧化氢(H₂O₂)诱导的人结肠腺癌Caco-2细胞氧化损伤的保护作用。
以H₂O₂诱导的Caco-2细胞氧化损伤为模型,测定不同浓度(0 - 80 μmol/L)的5 - n - 烷基间苯二酚预处理后Caco-2细胞的存活率,以确定最佳浓度范围。通过检测活性氧(ROS)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性水平,评估5 - n - 烷基间苯二酚对氧化损伤细胞的保护作用。
5 - n - 烷基间苯二酚在5 - 80 μmol/L浓度范围内对Caco-2细胞无明显毒性作用。600 μmol/L H₂O₂处理4小时后,细胞培养上清液和细胞中GSH-Px、SOD和CAT氧化酶活性均显著低于对照组(P<0.05)。添加不同浓度的5种烷基间苯二酚同系物不同程度地减轻了H₂O₂的损伤,与模型组相比,ROS水平和MDA含量降低,GSH-Px、SOD和CAT活性有一定程度恢复。
5 - n - 烷基间苯二酚可通过激活细胞内抗氧化酶活性和抑制脂质过氧化水平,保护Caco-2细胞免受H₂O₂诱导的氧化损伤。
