Smilacis Glabrae Rhizoma-Dioscoreae Spongiosae Rhizoma (SGR-DSR), a herb pair used in traditional Chinese medicine (TCM), has definite curative effect on the clinical treatment of gout. To screen and identify the anti-gout ingredients from SGR-DSR. In this study, an integration strategy combining ultrafiltration-high performance liquid chromatography-mass spectrometry (UF-HPLC-MS) and on-line HPLC-2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS)-MS were proposed and applied for the rapid screening and identification of anti-gout components (xanthine oxidase [XOD] inhibitors and free radical scavengers) from SGR-DSR. As a result, 35 compounds with both potential XOD inhibitor activity and free radical scavenging activity were screened from the herbal pair. Among them, 18 active components were verified by in vitro activity assays. Astilbin showed better XOD inhibition activity and ABTS scavenging ability with the inhibition rates of 73.76% and 81.56% at 80 µM, respectively. In addition, the molecular docking simulation calculation results showed that all the 18 components had good binding ability with XOD. This study indicated that herb pair with clear clinical efficacy of TCM was a promising source for anti-gout ingredients, and the integration screening strategy provides rapid and effective identification method of anti-gout ingredients from complex herb pairs.
绵萆薢-土茯苓药对(SGR-DSR)是一种用于中医临床治疗痛风有确切疗效的药对。旨在从SGR-DSR中筛选并鉴定抗痛风成分。本研究提出并应用超滤-高效液相色谱-质谱联用(UF-HPLC-MS)和在线HPLC-2,2'-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐(ABTS)-MS的集成策略,用于从SGR-DSR中快速筛选和鉴定抗痛风成分(黄嘌呤氧化酶[XOD]抑制剂和自由基清除剂)。结果,从该药对中筛选出35种同时具有潜在XOD抑制活性和自由基清除活性的化合物。其中,18种活性成分通过体外活性测定得到验证。落新妇苷在80 μM时表现出较好的XOD抑制活性和ABTS清除能力,抑制率分别为73.76%和81.56%。此外,分子对接模拟计算结果表明,这18种成分均与XOD具有良好的结合能力。本研究表明,具有明确临床疗效的中药药对是抗痛风成分的一个有前景的来源,且该集成筛选策略为从复杂药对中快速有效鉴定抗痛风成分提供了方法。
