Bacteriophages are seen as one of the most promising alternatives to antibiotic treatment in fight against drug resistant bacteria. However, as the interest in therapeutic potential of phages grew it became clear that even phages have their limitations. Narrow spectrum of activity, preference for bacteria in specific phase of metabolic activity or development of resistance mechanisms to phages may influence the efficacy of phage treatment. Therefore, in order to avoid those effects pairing bacteriophages with other antimicrobials is being researched. In our work we have paired staphylococcal phage vB_SauM-A with lactoferrin, a protein with proven antimicrobial activity and we have studied how this combination affected phage performance. We have observed that addition of 1 mg/mL of lactoferrin resulted in increased efficacy of plating of phage lysate. We have also noticed that it has influenced phage adsorption and life cycle kinetics on two out of four tested Staphylococcus aureus strains. Finally, in vivo experiment with Galleria mellonella showed that phage-lactoferrin mixture had higher therapeutic effect than the phage alone. Therefore, we assume that supplementation of phage cocktails with lactoferrin may increase the efficacy of the treatment. However, the subject requires more studies using different groups of phages and pathogens.
噬菌体被视为对抗耐药细菌的抗生素治疗最有前景的替代方法之一。然而,随着对噬菌体治疗潜力的兴趣增加,很明显即使是噬菌体也有其局限性。窄谱活性、对处于特定代谢活性阶段的细菌的偏好或对噬菌体抗性机制的发展可能会影响噬菌体治疗的效果。因此,为了避免这些影响,正在研究将噬菌体与其他抗菌剂配对。在我们的工作中,我们将葡萄球菌噬菌体vB_SauM-A与乳铁蛋白配对,乳铁蛋白是一种具有已证实抗菌活性的蛋白质,我们研究了这种组合如何影响噬菌体性能。我们观察到添加1mg/mL的乳铁蛋白导致噬菌体裂解物的平板接种效率提高。我们还注意到它对四种测试金黄色葡萄球菌菌株中的两种影响了噬菌体吸附和生命周期动力学。最后,用大蜡螟进行的体内实验表明,噬菌体-乳铁蛋白混合物比单独的噬菌体具有更高的治疗效果。因此,我们假设用乳铁蛋白补充噬菌体鸡尾酒可能会提高治疗效果。然而,该主题需要使用不同组的噬菌体和病原体进行更多研究。
