The immunoadsorbent 2,4-dinitrophenyl-amino-ethyl cellulose (DNP—AEC) was prepared by the reaction of dinitrofluorobenzene and aminoethyl cellulose in an alkaline medium. It was found to be an insoluble stable complex, specific for dinitro-phenyl antibody. The immunoadsorbent specifically retained a protein fraction (Fr. C) from pooled and individual human γ-globulins. This fraction was shown to behave like anti-DNP antibody when tested by equilibrium dialysis, fluorescent quenching and spectral shift analysis. The antibody activity was found to reside in the Fab fragment. The significance of finding naturally occuring anti-DNP antibodies in normal serum is discussed.
