Abstract
- Guanase has been purified 200-fold in 20% yield from the supernatant fraction of rabbit-liver homogenates, by using ammonium sulphate fractionation, calcium phosphate-gel adsorption and chromatography on DEAE-cellulose and Sephadex G-200. 2. K(m) with guanine as substrate at the optimum pH of 7.7 was found to be 1.05x10(-5)m. Q(10) was 1.4 between 23 degrees and 48 degrees . 3. Substrate activity and pH optima of compounds related to guanine have been studied. 8-Azaguanine, 1-methylguanine, thioguanine and 1-methylthioguanine are all substrates.

- 利用硫酸铵分级分离、磷酸钙凝胶吸附以及在二乙氨基乙基纤维素和葡聚糖凝胶G - 200上进行色谱分离，从兔肝匀浆的上清液部分中以20%的产率将鸟嘌呤酶纯化了200倍。2. 在最佳pH值7.7时，以鸟嘌呤为底物的米氏常数（K(m)）为1.05×10⁻⁵摩尔/升。在23摄氏度至48摄氏度之间，温度系数（Q(10)）为1.4。3. 对与鸟嘌呤相关的化合物的底物活性和最适pH值进行了研究。8 - 氮杂鸟嘌呤、1 - 甲基鸟嘌呤、硫代鸟嘌呤和1 - 甲基硫代鸟嘌呤均为底物。
