Abstract

The kinetic parameters of azoalbumin hydrolysis by alkaline proteinase from Bacillus subtilis were determined to be Km = 1.2 . 10(-3) M, kcat = 1.5 sec-1 according to the method of Lainuiwer-Berk and Km = 4 . 10(-3) M, kcat = 0.5 sec-1 from the analysis of the entire kinetic curve. It was found that pH optimum of subtilizin hydrolysis of various substrates and the shape of the curve depended on the substrate nature.

根据莱尼韦尔-伯克方法,枯草芽孢杆菌碱性蛋白酶水解偶氮白蛋白的动力学参数测定为Km = 1.2×10⁻³ M,kcat = 1.5秒⁻¹;而通过对整个动力学曲线分析得出的参数为Km = 4×10⁻³ M,kcat = 0.5秒⁻¹。研究发现,嗜热栖热菌蛋白酶对各种底物水解的最适pH值以及曲线形状取决于底物的性质。
