Immune tolerance has an antigen-directed specificity analogous to that of immune responsiveness. Studies have shown the direction of response-immunization or tolerization-is often dependent on the physical form and the dose of antigen, and that the two major lymphocyte classes, B cells and T cells, have different tolerance requirements. The critical relationships of these findings to autoimmunity are explored.
