To examine the relationship among the expression of protooncogenes such as c-fos and c-myc, Fas antigen, Fas ligand, and apoptosis in the synovial tissue of patients with rheumatoid arthritis (RA).
The expression of c-fos, c-myc, Fas antigen, and Fas ligand was examined in synovial tissues of 6 patients with RA and 4 with osteoarthritis (OA) using in situ reverse transcriptase (RT) assay and immunohistochemical staining. Apoptosis was detected by TUNEL method in situ.
Expression of protooncogenes, c-fos, and c-myc was detected in all samples from patients with RA, but in only a few cells of OA synovium. 30 to 90% of cells in RA synovium positive for these protooncogenes also coexpressed Fas antigen. Fas positive cells in RA synovium underwent apoptosis to a significant degree. Fas ligand mRNA was detected only in mononuclear cells in RA synovium.
The expression of protooncogenes is closely related to Fas mediated apoptosis in RA synoviocytes.

采用原位逆转录酶(RT)法和免疫组化染色检测6例RA患者及4例骨关节炎(OA)患者滑膜组织中c-fos、c-myc、Fas抗原和Fas配体的表达。采用原位TUNEL法检测细胞凋亡。
RA患者所有样本均检测到原癌基因c-fos和c-myc的表达,而OA滑膜组织中只有少数细胞表达。这些原癌基因在RA滑膜中30%至90%的阳性细胞也共表达Fas抗原。RA滑膜中的Fas阳性细胞发生了显著程度的凋亡。Fas配体mRNA仅在RA滑膜的单核细胞中检测到。
原癌基因的表达与RA滑膜细胞中Fas介导的细胞凋亡密切相关。
