To observe the changes of interleukin-1 (IL-1), nitric oxide (NO) and nitric oxide synthase (NOS) in mice with oleic acid-induced acute lung injury (ALI) and the protective effects of interleukin-1 receptor antagonist (IL-1ra).
Male Kunming mice were divided into control, oleic acid and IL-1ra groups. The control group mice were injected saline; the oleic acid group mice were injected oleic acid (0.2 ml/kg): and the IL-1ra group mice were injected the IL-1ra (20 mg/kg). Lung index, lung wet-to-dry weight ratio, and total protein, cell analysis, nitric oxide measurement, NOS activity in BALF, lung pathology examination were made after an hour of administration of drug.
Preadministration of IL-1ra to the mouse with ALI decreased the lung index, lung wet-to-dry weight ratio and leakage of protein from pulmonary capillary, elevated PaO2, and attenuated lung histologic injury. It was found that in bronchoalveolar lavage fluid (BALF), NO amount and lung NOS activity increased in oleic acid group, BALF NO amount and lung NOS activity decreased obviously after given IL-1ra.
This study demonstrated the protective effect of IL-1ra on oleic acid-induced lung injury, NO may participate in the pathological process of lung injury.
观察油酸诱导的急性肺损伤(ALI)小鼠体内白细胞介素-1(IL-1)、一氧化氮(NO)和一氧化氮合酶(NOS)的变化以及白细胞介素-1受体拮抗剂(IL-1ra)的保护作用。
雄性昆明小鼠分为对照组、油酸组和IL-1ra组。对照组小鼠注射生理盐水;油酸组小鼠注射油酸(0.2 ml/kg);IL-1ra组小鼠注射IL-1ra(20 mg/kg)。给药1小时后进行肺指数、肺湿重与干重比值、总蛋白、细胞分析、一氧化氮测定、支气管肺泡灌洗液(BALF)中NOS活性测定及肺病理检查。
给ALI小鼠预先注射IL-1ra可降低肺指数、肺湿重与干重比值以及肺毛细血管蛋白渗漏,提高动脉血氧分压(PaO2),减轻肺组织学损伤。发现油酸组支气管肺泡灌洗液(BALF)中NO含量和肺NOS活性升高,给予IL-1ra后BALF中NO含量和肺NOS活性明显降低。
本研究证明了IL-1ra对油酸诱导的肺损伤具有保护作用,NO可能参与肺损伤的病理过程。
