Two ophthalmic solutions of 0.3% ciprofloxacin eye drops are available in Turkey: Ciloxan and Siprogut. A previous study by the same authors was the first to report vitreous penetration of ciprofloxacin-containing eye drops. The aim of the present study was to compare the levels of drug found in the subretinal fluid by the two products following local administration.
Forty-three patients undergoing conventional retinal detachment surgery received either Ciloxan (22 patients) or Siprogut (21 patients). Beginning 6 h before surgery, two drops of solution were instilled onto the operative eye every 30 min for the first 3 h and then hourly for the next 3 h. Subretinal fluid samples were collected 30 min after administration of the last dose and were assayed for ciprofloxacin levels using a method involving high-performance liquid chromatography with fluorometric detection.
The minimum and maximum subretinal fluid concentrations measured were 0.11 microg/mL and 0.65 microg/mL, respectively, with Ciloxan, and 0.08 microg/mL and 0.62 microg/mL, respectively wth Siprogut. There was no statistical difference between the subretinal fluid ciprofloxacin levels of the two products. The subretinal fluid drug evels attained by both products were below the minimum inhibitory concentrations of common ocular pathogens.
Ciloxan and Siprogut can penetrate subretinal fluid. The ocular bioavailability of ciprofloxacin after local administration is equivalent for both pharmaceutical products.
在土耳其有两种0.3%的环丙沙星眼药水可供使用:Ciloxan和Siprogut。同一组作者之前的一项研究首次报道了含环丙沙星眼药水的玻璃体穿透情况。本研究的目的是比较这两种产品局部给药后视网膜下液中的药物水平。
43例行传统视网膜脱离手术的患者分别使用Ciloxan(22例)或Siprogut(21例)。在手术前6小时开始,最初3小时每30分钟向术眼滴入两滴溶液,然后在接下来的3小时每小时滴入两滴。在最后一剂给药后30分钟采集视网膜下液样本,采用高效液相色谱荧光检测法测定环丙沙星水平。
使用Ciloxan时,测得的视网膜下液最低和最高浓度分别为0.11微克/毫升和0.65微克/毫升,使用Siprogut时分别为0.08微克/毫升和0.62微克/毫升。两种产品的视网膜下液环丙沙星水平之间无统计学差异。两种产品达到的视网膜下液药物水平均低于常见眼部病原体的最低抑菌浓度。
Ciloxan和Siprogut均可穿透视网膜下液。两种药品局部给药后环丙沙星的眼部生物利用度相当。
