Abstract

AIM

To investigate the effect of cytokines on the liver necrosis.METHODS: rIL (interleukin)-1, rIL-6, rIFN (interferon), rTNF (tumor necrosis factor) -alpha with or without D-galactosamine (D-GAL) were injected into the abdominal cavity of mice separately.ALT, TBIL (total bilirubin) and histological changes were observed.RESULTS: There was no effect on hepatocyte of normal mice after injection of rIL-1, rIL-6, rIFN alone or together. The serum total bilirubin (TBIL) and liver necrosis of mice increased after rTNF-alpha, rIL-6 or rIFN were used separately with D-GAL. The TBIL level (&mgr;mol/L)was 46.19 plus minus 10.62, 44.55 plus minus 12.9 and 41.94 plus minus 14.9, higher than that caused by D-GAL alone (TBIL, 26.67&mgr;mol/L plus minus 11.14&mgr;mol/L). The serum TBIL of mice and the degree of liver necrosis increased after injection of IL-1, IL-6 with D-GAL and rTNF-alpha.CONCLUSION: Cytokines, like IL-1, IL-6, IFN&agr and TNF-&agr;joined in the process of hepatocyte necrosis.They can enhance the degree of liver necrosis induced by D-GAL.

目的

研究细胞因子对肝坏死的影响。方法:将重组白细胞介素-1(rIL-1)、重组白细胞介素-6(rIL-6)、重组干扰素(rIFN)、重组肿瘤坏死因子-α(rTNF-α)单独或与D-半乳糖胺(D-GAL)一起注入小鼠腹腔。观察谷丙转氨酶(ALT)、总胆红素(TBIL)及组织学变化。结果:单独或联合注射rIL-1、rIL-6、rIFN对正常小鼠肝细胞无影响。rTNF-α、rIL-6或rIFN与D-GAL单独使用后,小鼠血清总胆红素(TBIL)及肝坏死增加。TBIL水平(μmol/L)分别为46.19±10.62、44.55±12.9和41.94±14.9,高于单独使用D-GAL时(TBIL,26.67μmol/L±11.14μmol/L)。注射IL-1、IL-6与D-GAL及rTNF-α后,小鼠血清TBIL及肝坏死程度增加。结论:细胞因子如IL-1、IL-6、IFNα和TNF-α参与肝细胞坏死过程,可增强D-GAL诱导的肝坏死程度。
