To quest for a simple and convenient assay for the diagnosis of schistosomiasis.
Three monoclonal antibodies against different epitopes of antigens of S. japonicum were used to develop a dot immunogold filtration assay for detection of schistosomal antigen-antibody reaction on the membrane during the filtration process. The results could be read with the naked eye within a few minutes.
The sensitivity of the SEA assay was 16 ng/ml. In 69 serum samples from patients with chronic schistosomiasis, the positive rate and specificity of this assay were 60.8% and 95.2%, respectively. Dot-ELISA was used in detecting 137 serum samples from patients with chronic schistosomiasis, the positive rate and specificity were 54.7% and 94.6%, respectively. When sandwich-ELISA was used in detecting 118 serum samples from patients with chronic schistosomiasis, the positive rate and specificity were 61.9% and 95.7%, respectively.
The sensitivity and specificity of DIFA were similar to those of ELISA, but DIFA required less time and no special equipment.
探寻一种简单便捷的血吸虫病诊断检测方法。
使用三种针对日本血吸虫抗原不同表位的单克隆抗体,建立斑点免疫金渗滤法,用于检测渗滤过程中膜上的血吸虫抗原 - 抗体反应。结果可在数分钟内用肉眼读取。
SEA检测法的灵敏度为16 ng/ml。在69份慢性血吸虫病患者血清样本中,该检测法的阳性率和特异性分别为60.8%和95.2%。采用斑点ELISA检测137份慢性血吸虫病患者血清样本,阳性率和特异性分别为54.7%和94.6%。采用夹心ELISA检测118份慢性血吸虫病患者血清样本,阳性率和特异性分别为61.9%和95.7%。
斑点免疫金渗滤法(DIFA)的灵敏度和特异性与ELISA相似,但DIFA所需时间更短且无需特殊设备。
