To compare the chemical composition and bioactivity of polypeptides(PPs) isolated from velvet antlers of sika deer (Cervus nippon Temminck) and red deer (Cervus elaphus Linnaeus).
The two kind of polypeptides were isolated from the above mentioned velvet antlers with same technology. The chemical composition was determined using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and MALDI-TOF mass spectrometry. Stimulant activity of cells proliferation was measured by [3H] TdR incorporation into DNA.
The graphs of SDS-PAGE and MALDI-TOF MS of velvet antler polypeptides (VAPPs) from Chinese and New zealand red deer were very similar, but there were obvious difference in respect of graph between sika deer and red deer. VAPPs 25-50 mg.L-1 showed marked proliferation-promoting activity for rabbit costed chondrocytes, either sika deer or red deer. However, the activity of sika deer VAPPs 12.5 mg.L-1 for epidermal cells was weaker than that of red deer (12.5 mg.L-1).
The chemical property and bioactivity of VAPPs from sika deer and red deer are significantly different.
比较从梅花鹿(Cervus nippon Temminck)和马鹿(Cervus elaphus Linnaeus)鹿茸中分离得到的多肽(PPs)的化学组成和生物活性。
采用相同技术从上述鹿茸中分离两种多肽。使用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)和基质辅助激光解吸电离飞行时间质谱(MALDI-TOF)测定化学组成。通过[3H]胸腺嘧啶核苷掺入DNA来测量细胞增殖的刺激活性。
中国和新西兰马鹿鹿茸多肽(VAPPs)的SDS-PAGE图谱和MALDI-TOF质谱图非常相似,但梅花鹿和马鹿的图谱存在明显差异。25 - 50 mg.L-1的VAPPs对兔肋软骨细胞显示出显著的促增殖活性,无论是梅花鹿还是马鹿的。然而,12.5 mg.L-1的梅花鹿VAPPs对表皮细胞的活性弱于马鹿(12.5 mg.L-1)。
梅花鹿和马鹿的VAPPs的化学性质和生物活性存在显著差异。
