To investigate the therapeutic effect of 125IUdR on gliomas.
By means of growth-curve, clonogenic formation experiment and MTT assay, the inhibitive effect of 125IUdR on the proliferation of C6 cells was studied in vitro. Wistar rats with intracerebral C6 gliomas were used to verify the therapeutic efficacy of 125IUdR in vivo.
C6 monolayer cells were efficiently inhibited by 125IUdR in a time-and-dose-dependent manner. In the MTT assay, after treament with 150 kBq/ml 125IUdR for 5 days, the inhibition rate reached 93.06%. In murine transplantable tumor, 125IUdR had significant therapeutic effect on rats bearing solid tumor glioma C6. After treatment with 125IUdR for 5 days, the tumor weight of experiment group was lower than that of blank group and control group (P<0.01). The median survival of animals treated with 125IUdR (27 days) was markedly longer than that of control group (9 days) (P<0.01). Na 125I and 127IUdR showed little inhibitive effect on the proliferation of C6 cells in vitro and in vivo.
125IUdR can markedly inhibit the growth of Glioma cell line C6. 125IUdR has potential for the treatment of malignant brain tumor.
探讨125IUdR对胶质瘤的治疗效果。
通过生长曲线、克隆形成实验和MTT法,体外研究125IUdR对C6细胞增殖的抑制作用。采用脑内接种C6胶质瘤的Wistar大鼠在体内验证125IUdR的治疗效果。
125IUdR能以时间和剂量依赖的方式有效抑制C6单层细胞。在MTT法中,用150 kBq/ml 125IUdR处理5天后,抑制率达到93.06%。在小鼠可移植肿瘤中,125IUdR对荷实体瘤胶质瘤C6的大鼠有显著治疗效果。用125IUdR处理5天后,实验组肿瘤重量低于空白组和对照组(P<0.01)。125IUdR处理动物的中位生存期(27天)明显长于对照组(9天)(P<0.01)。125I钠和127IUdR在体外和体内对C6细胞增殖的抑制作用很小。
125IUdR能显著抑制胶质瘤细胞系C6的生长。125IUdR具有治疗恶性脑肿瘤的潜力。
