Abstract

OBJECTIVE

To establish a method for determination of galanthamine in Lycoris radiata.

METHOD

HPLC separation was carried on a column of ODS (4.6 mm x 150 mm, 5 microm), with the mobile phase of phosphate buffer (pH = 3-4)-methanol (93:7) at the flow rate of 1.0 mL x min(-1), and the detection wavelength was set at 289 nm.

RESULT

Galanthamine revealed linearity within the range of 3-30 microg x mL(-1) (r = 0.9997), the detection limit was 0.3 ng. The average recovery was 99.5% (RSD = 0.5%).

CONCLUSION

The method is easy to operate and the results of the determination are accurate, it can be used to evaluate the quality of L. radiata.

目的

建立测定石蒜中加兰他敏的方法。

方法

采用ODS柱(4.6 mm×150 mm,5μm)进行高效液相色谱分离,流动相为磷酸盐缓冲液(pH = 3 - 4)-甲醇(93:7),流速为1.0 mL·min⁻¹,检测波长设定为289 nm。

结果

加兰他敏在3 - 30μg·mL⁻¹范围内呈线性关系(r = 0.9997),检测限为0.3 ng。平均回收率为99.5%(相对标准偏差RSD = 0.5%)。

结论

该方法操作简便,测定结果准确,可用于评价石蒜的质量。
