To investigate the molecular mechanisms and effective target points of lipid-lowering drug, Rhizoma Curcumae Longae, and study the effect of curcumin on the expression of low density lipoprotein (LDL) receptors in macrophages in mice.
Macrophages in mice were treated with curcumin, which was purified from the ethanolly extraction of Rhizoma Curcumae Longae for 24 h. The LDL receptors expressed in the macrophages were determined by enzyme-linked immunosorbent assay (ELISA) and assay of DiI labeled LDL uptake by flow cytometer.
It was found for the first time that 10 micromol/L-50 micromol/L curcumin could obviously up-regulate the expression of LDL receptor in macrophages in mice, and a dose-effect relationship was demonstrated.
One of the lipid-lowering mechanisms of traditional Chinese medicine, Rhizoma Curcumae Longae, was completed by the effect of curcumin through the up-regulation of the expression of LDL receptor.
探讨降脂药物姜黄的分子作用机制及有效靶点,研究姜黄素对小鼠巨噬细胞低密度脂蛋白(LDL)受体表达的影响。
用从姜黄乙醇提取物中纯化得到的姜黄素处理小鼠巨噬细胞24小时。采用酶联免疫吸附测定(ELISA)法和流式细胞仪检测DiI标记的LDL摄取量来测定巨噬细胞中表达的LDL受体。
首次发现10微摩尔/升-50微摩尔/升姜黄素能明显上调小鼠巨噬细胞中LDL受体的表达,并呈现剂量-效应关系。
中药姜黄的降脂机制之一是通过姜黄素上调LDL受体的表达来实现的。
