To study the protective effects of hydroxyethylpuerarin against the injury of astrocytes induced by hydrogen peroxide (H2O2).
Experiments were performed with cells from passage 4. Plasma membrane integrity was measured by lactate dehydrogenase (LDH) release. The occurrence of apoptosis was measured by flow cytometry. The glutamate uptake of astrocytes was studied with [3H]-glutamate incorporation. Intracellular superoxide dismutase (SOD) activity and malondialdehyde (MDA) level were assessed by automatic biochemistry analyzer.
Compared with H2O2 injured group, the occurrence of apoptosis, levels of LDH release and intracellular MDA of astrocytes reduced in hydroxyethylpuerarin pre-treated groups, but the glutamate uptake and intracellular SOD activity of astrocytes increased.
Hydroxyethylpuerarin could reduce the occurrence of apoptosis and improve neurotrophic function of astrocytes, which may be related with its antioxidant effects during oxidative stress.
研究羟乙基葛根素对过氧化氢(H2O2)诱导的星形胶质细胞损伤的保护作用。
采用第4代细胞进行实验。通过乳酸脱氢酶(LDH)释放检测细胞膜完整性。采用流式细胞术检测细胞凋亡情况。用[3H] - 谷氨酸掺入法研究星形胶质细胞对谷氨酸的摄取。通过自动生化分析仪评估细胞内超氧化物歧化酶(SOD)活性和丙二醛(MDA)水平。
与H2O2损伤组相比,羟乙基葛根素预处理组星形胶质细胞的凋亡发生率、LDH释放水平和细胞内MDA水平降低,但星形胶质细胞的谷氨酸摄取和细胞内SOD活性增加。
羟乙基葛根素可减少星形胶质细胞凋亡的发生并改善其神经营养功能,这可能与其在氧化应激期间的抗氧化作用有关。
