To develop a quantitative analytical procedure of scopolamine and atropine in Flos Daturae using RP-HPLC.
The two alkaloids were separated on a Hypersil BDS C18 column (4.6 mm x 250 mm, 5 microm) with a mobile phase of 0.02 mol x L(-1) sodium acetate buffer (containing 0.02% triethanolamine and the pH was adjusted to 6.0 with acetic acid)-methanol (60:40) and a detection wavelength of 215 nm. The flow rate was 1.0 mL x min(-1) and the column temperature was maintained at room temperature.
The mean recovery was (99.6 +/- 1.8)% for scopolamine and (100.4 +/- 1.5)% for atropine.
This method was simple, accurate and sensitive.
建立用反相高效液相色谱法测定洋金花中莨菪碱和阿托品含量的定量分析方法。
采用Hypersil BDS C18柱(4.6 mm×250 mm,5μm),以0.02 mol·L⁻¹醋酸钠缓冲液(含0.02%三乙醇胺,用醋酸调pH至6.0)-甲醇(60∶40)为流动相,检测波长为215 nm。流速为1.0 mL·min⁻¹,柱温保持在室温。
莨菪碱平均回收率为(99.6±1.8)%,阿托品平均回收率为(100.4±1.5)%。
该方法简便、准确、灵敏。
