To study characteristic fingerprints of the fat-soluble components of different Marsdenia tenacissima samples by GC and GC-MS.
The sample was split in the 280 degrees C injection port, with 10:1 split ratio, and separated on a fused silica capillary column (DB-5, 30 m x 0.25 mm I. D., 0.25 microm film). The temperature program was 70 degrees C for 5 min, 8 degrees C x min(-1) to 200 degrees C, then 5 degrees C x min-1 to 260 degrees C, 260 degrees C for 10 min. The high pure nitrogen was used as a carrier gas, The FID temperature was 300 degrees C.
There are 15 common peaks in the GC fingerprints of M. tenacissima and they are identified by GC-MS. The analyses on cluster and similarity degree of the finger the crude drugs from various habitats have been performed.
The method has good repeatability. The GC fingerprint combined with the HPLC fingerprint of M. tenacissim can be used to evaluate its quality integrally.
采用气相色谱(GC)和气相色谱-质谱联用(GC-MS)技术研究不同萝摩样品脂溶性成分的特征指纹图谱。
样品在280℃进样口进行分流,分流比为10:1,在熔融石英毛细管柱(DB-5,30 m×0.25 mm内径,0.25μm膜厚)上分离。程序升温条件为:70℃保持5 min,以8℃/min升至200℃,再以5℃/min升至260℃,260℃保持10 min。高纯氮气为载气,氢火焰离子化检测器(FID)温度为300℃。
萝摩GC指纹图谱中有15个共有峰,并通过GC-MS进行了鉴定。对不同产地药材指纹图谱进行了聚类分析和相似度分析。
该方法重复性良好。萝摩的GC指纹图谱与高效液相色谱(HPLC)指纹图谱相结合可用于整体评价其质量。
