Naturally occurring autoantibodies against native DNA (nDNA) in SLE sera showed diverse antigen binding characteristics. The antibodies isolated by affinity chromatography using nDNA linked to Sepharose 4B exhibited specificity towards nDNA and showed strong reactivity with DNA-psoralen photoadduct by direct binding assay and competitive ELISA. The anti-DNA antibody belong to both IgG and IgM immunoglobulin classes and their ratio was 5:1. The possible significance of altered conformation of nDNA in the etiology of SLE has been discussed.
系统性红斑狼疮(SLE)血清中天然存在的抗双链DNA(nDNA)自身抗体表现出多样的抗原结合特性。通过使用与琼脂糖4B偶联的nDNA进行亲和层析分离得到的抗体对nDNA具有特异性,并且通过直接结合试验和竞争性酶联免疫吸附测定(ELISA)显示出与DNA-补骨脂素光加合物具有强烈反应性。抗DNA抗体属于IgG和IgM两类免疫球蛋白,其比例为5:1。文中讨论了nDNA构象改变在SLE病因学中的可能意义。
