The aims of this study were to investigate (1) the matrix metalloproteinase-1 (MMP-1) promoter polymorphisms in severe chronic periodontitis (CP), (2) the relationship of periodontal therapy outcome with these genotypes, and (3) the gingival crevicular fluid (GCF) MMP-1 levels-MMP-1 genotype correlation.
Genomic DNA was obtained from the peripheral blood of 102 patients with severe CP and 98 periodontally healthy subjects. MMP-1 -519A/G and -1607 1G/2G polymorphisms were determined by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Fifty-eight CP patients received non-surgical periodontal therapy and were followed for 6 months. Clinical periodontal parameters and GCF samples were collected at baseline and at 6 months. GCF MMP-1 levels were analysed by enzyme-linked immunosorbent assay (ELISA).
The distribution of MMP-1 genotypes did not significantly differ between the study groups. On the other hand, the -1607 2G allele frequency of severe CP patients was higher than that of healthy subjects. MMP-1 -519G allele carriers had higher GCF MMP-1 levels and percentage of sites with 4-6 mm clinical attachment level (CAL) compared with AA genotypes after non-surgical periodontal therapy (p<0.05).
These data suggest that the -1607 2G polymorphic allele of the MMP-1 gene could be associated with susceptibility to severe CP in the Turkish population. It seems that -519AG and GG genotypes could play a role in the outcome of periodontal therapy.
本研究的目的是调查(1)重度慢性牙周炎(CP)中基质金属蛋白酶-1(MMP-1)启动子多态性,(2)牙周治疗结果与这些基因型的关系,以及(3)龈沟液(GCF)中MMP-1水平与MMP-1基因型的相关性。
从102例重度CP患者和98例牙周健康受试者的外周血中获取基因组DNA。采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法测定MMP-1 -519A/G和-1607 1G/2G多态性。58例CP患者接受了非手术牙周治疗,并随访6个月。在基线和6个月时收集临床牙周参数和GCF样本。通过酶联免疫吸附测定(ELISA)分析GCF中MMP-1水平。
研究组之间MMP-1基因型的分布没有显著差异。另一方面,重度CP患者的-1607 2G等位基因频率高于健康受试者。非手术牙周治疗后,与AA基因型相比,MMP-1 -519G等位基因携带者的GCF中MMP-1水平更高,且临床附着水平(CAL)为4-6 mm的位点百分比更高(p<0.05)。
