To optimize the prescription and preparation of aconitine liposomes.
Aconitine liposomes were prepared by ethanol injection method. The encapsulation efficiency was taken as inspection target and the preparation of liposomes were optimized by orthogonal design. HPLC was used to measure the encapsulation efficiency.
The best prescription was aconitine/ lecithin/cholesterol( 1: 15: 5)with phosphate buffer solution (pH 7. 4) as hydration media. The hydration temperature was 60 degrees C.
The optimized formulation of aconitine liposomes is reasonable in prescription, practicable in technology, high in encapsulation efficiency and good in stability.
优化乌头碱脂质体的处方和制备工艺。
采用乙醇注入法制备乌头碱脂质体。以包封率为考察指标,采用正交设计法优化脂质体制备工艺。采用高效液相色谱法测定包封率。
最佳处方为乌头碱/卵磷脂/胆固醇(1:15:5),以磷酸盐缓冲液(pH 7.4)为水化介质,水化温度为60℃。
优化后的乌头碱脂质体处方合理,工艺可行,包封率高,稳定性好。
