Polymorphonuclear neutrophils (PMNs) act as a first line of defense against bacterial infections. Recent studies indicated that PMNs could act as antigen-presenting cells. The aim of this study was to detect the expression of major histocompatibility complex (MHC) class II and certain molecules and receptors (CD83, CD64, and CD14) on the surface of PMNs stimulated with interleukin (IL)-15.
PMNs were isolated from heparinized whole blood of healthy volunteers. Cells were incubated for 48 h at 37 degrees C in a humidified incubator with 5% carbon dioxide. After incubation for 48 h with and without IL-15, surface molecules were assayed by cytofluorometry.
Stimulation with IL-15 produced an increase in the expression of MHC class II and upregulation of the costimulatory molecules CD83, the immunoglobulin G receptor CD64, and the lipopolysaccharide receptor CD14 on the surface of PMNs.
The expression of MHC class II, CD83, CD64, and CD14 indicates that PMNs may be involved in both innate and acquired immunity.
多形核中性粒细胞(PMNs)是抵御细菌感染的第一道防线。近期研究表明,PMNs可作为抗原呈递细胞。本研究旨在检测白细胞介素(IL)-15刺激后PMNs表面主要组织相容性复合体(MHC)II类分子以及某些分子和受体(CD83、CD64和CD14)的表达情况。
从健康志愿者的肝素化全血中分离出PMNs。细胞在含5%二氧化碳的加湿培养箱中于37℃孵育48小时。在有和无IL-15的情况下孵育48小时后,通过细胞荧光测定法检测表面分子。
IL-15刺激使PMNs表面MHC II类分子的表达增加,共刺激分子CD83、免疫球蛋白G受体CD64和脂多糖受体CD14上调。
MHC II类分子、CD83、CD64和CD14的表达表明PMNs可能参与天然免疫和获得性免疫。
