The conformation of a subset of phosphorylated serines or threonines preceding proline motifs is regulated by the prolyl isomerase Pin1. Pin1 plays a critical role in oncogenesis. The aim of this study is to explore the relationship between the expression of Pin1 and clinicopathological factors in squamous cell carcinoma and adenocarcinoma of the lung, and to analyze the correlation between Pin1 and β-catenin.
The expression of Pin1 and β-catenin proteins was detected in 69 lung cancer cases by immunohistochemical SP method, and in 30 fresh lung samples by Western blot.
Immunohistochemically, the overexpression of Pin1 and β-catenin in lung cancer was 78.3% (54/69) and 63.8% (44/69), respectively. The expression of Pin1 and β-catenin was not related to age, sex, histological classification, differentiation, lymph node metastasis and pTNM stages. There was a positive correlation between overexpression of Pin1 and aberrant β-catenin expression (P < 0.05). Western blot results showed that the expression of Pin1 and β-catenin in lung cancer tissues was significantly higher than that of paracancerous lung tissues (P < 0.05).
Pin1 is overexpressed in squamous cell carcinoma and adenocarcinoma of the lung and may play a critical role in oncogenesis of lung cancer. Overexpression of Pin1 might contribute to the upregulation of β-catenin and it may be one of the pathways for Pin1 to work.
脯氨酸基序之前的磷酸化丝氨酸或苏氨酸亚组的构象受脯氨酰异构酶Pin1调控。Pin1在肿瘤发生中起关键作用。本研究旨在探讨Pin1表达与肺鳞状细胞癌和腺癌临床病理因素之间的关系,并分析Pin1与β-连环蛋白之间的相关性。
采用免疫组织化学SP法检测69例肺癌病例中Pin1和β-连环蛋白蛋白的表达,并采用蛋白质印迹法检测30例新鲜肺组织样本中的表达。
免疫组织化学结果显示,肺癌中Pin1和β-连环蛋白的过表达率分别为78.3%(54/69)和63.8%(44/69)。Pin1和β-连环蛋白的表达与年龄、性别、组织学分类、分化程度、淋巴结转移及pTNM分期无关。Pin1过表达与β-连环蛋白异常表达呈正相关(P<0.05)。蛋白质印迹结果显示,肺癌组织中Pin1和β-连环蛋白的表达明显高于癌旁肺组织(P<0.05)。
Pin1在肺鳞状细胞癌和腺癌中过表达,可能在肺癌发生中起关键作用。Pin1过表达可能导致β-连环蛋白上调,这可能是Pin1发挥作用的途径之一。
