Abstract

OBJECTIVE

To analyze the allele frequencies and polymorphism of human leukocyte antigens (HLA) -A, B, Cw, DRB1 and DQB1 between donors-recipients on high-resolution typing; and to analyze the matching and mismatching proportion between donors and recipients.

METHODS

HLA high-resolution types were determined by sequence based typing (SBT), sequence specific oligonucleotide probe (SSOP) and sequence specific primer (SSP) on 2540 unrelated Chinese Han individuals including 1168 recipients and 1372 donors, then statistical analyses were carried out.

RESULTS

Forty-four HLA-A alleles were detected, and among them the frequencies of A1101, A2402, A0201, A0207, A3303, A0206 and A3001 exceeded 0.05, and accounted for 80.4%. Eighty-one HLA-B alleles were detected, and the frequencies of B4001, B4601, B5801, B1302 and B5101 exceeded 0.05, and accounted for 43.0% of total. There were 44 HLA-Cw alleles, among them the frequencies of Cw0702, Cw0102, Cw0304, Cw0801, Cw0602, Cw0303, Cw0302 and Cw0401 exceeded 0.05, and were 80.3% of total. There were 61 HLA-DRB1 alleles, the frequencies of DRB10901, DRB11501, DRB11202, DRB10803, DRB10701, DRB10405, DRB10301 and DRB11101 exceeded 0.05, and were 70.1% of total. Finally, 22 HLA-DQB1 alleles were detected, the frequencies of DQB10301, DQB10303, DQB10601, DQB10602, DQB10202, DQB10302, DQB10401, DQB10502 and DQB1*0201 exceeded 0.05, and they were 87.4% of total. All the five loci were of heterozygote deficiency. The HLA-A, B and DRB1 loci conformed to Hardy-Weinberg equilibrium (HWE) (P > 0.05); but HLA-Cw and HLA-DQB1 loci did not (P < 0.05). Except several particular genotypes, all the five loci conformed to HWE. After comparing data between donors and recipients, only 22.4% of recipients found HLA matched donors (10/10); 24.6% of recipients found single HLA allele mismatched donors (9/10); 26.3% of recipients had two HLA alleles mismatched donors (8/10).

CONCLUSION

The characteristics of allele frequencies and polymorphism of HLA-A, B, Cw, DRB1 and DQB1 on high-resolution typing in Chinese Han population is valuable for donor searching in unrelated hematopoietic stem cell transplantation, and it provides genetic basis for donor registry and usage of donor resource for Chinese Marrow Donor Program.

采用基于序列的分型(SBT)、序列特异性寡核苷酸探针(SSOP)和序列特异性引物(SSP)对2540例中国汉族无关个体进行HLA高分辨率分型,其中包括1168例受者和1372例供者,然后进行统计分析。

结果

共检测到44个HLA-A等位基因,其中A1101、A2402、A0201、A0207、A3303、A0206和A3001的频率超过0.05,占80.4%。共检测到81个HLA-B等位基因,其中B4001、B4601、B5801、B1302和B5101的频率超过0.05,占总数的43.0%。共检测到44个HLA-Cw等位基因,其中Cw0702、Cw0102、Cw0304、Cw0801、Cw0602、Cw0303、Cw0302和Cw0401的频率超过0.05,占总数的80.3%。共检测到61个HLA-DRB1等位基因,其中DRB10901、DRB11501、DRB11202、DRB10803、DRB10701、DRB10405、DRB10301和DRB11101的频率超过0.05,占总数的70.1%。最后,共检测到22个HLA-DQB1等位基因,其中DQB10301、DQB10303、DQB10601、DQB10602、DQB10202、DQB10302、DQB10401、DQB10502和DQB1*0201的频率超过0.05,占总数的87.4%。所有五个位点均存在杂合子缺失。HLA-A、B和DRB1位点符合Hardy-Weinberg平衡(HWE)(P>0.05);但HLA-Cw和HLA-DQB1位点不符合(P<0.05)。除了几种特殊基因型外,所有五个位点均符合HWE。比较供受者数据后,仅22.4%的受者找到HLA匹配的供者(10/10);24.6%的受者找到单个HLA等位基因不匹配的供者(9/10);26.3%的受者有两个HLA等位基因不匹配的供者(8/10)。

结论

中国汉族人群高分辨率分型下HLA-A、B、Cw、DRB1和DQB1的等位基因频率及多态性特征对于无关造血干细胞移植供者的寻找具有重要价值,为中国造血干细胞捐献者资料库的供者登记及供者资源的利用提供了遗传依据。
