Abstract

OBJECTIVE

To understand the impacts of anthropogenic activities on structure and composition of bacterial communities and to evaluate how bacterial communities respond to environmental gradients at coastal sediments.

METHODS

The diversity of bacterial communities in sediments from tourist and mariculture zones at coastal area of Dalian Changshan Islands was assessed using terminal restriction fragment length polymorphism (t-RFLP) and denaturing gradient gel electrophoresis (DGGE) approaches. Meanwhile, 16S rRNA clone library was constructed to reveal the composition and structure of bacterial communities in the most seriously polluted site (D4).

RESULTS

There were much higher values of richness, Shannon-wiener and evenness index at D4 site by the analysis of terminal restriction fragments (t-RFs). The clustering result on the t-RFs areas and DGGE patterns showed that the bacterial diversity of tourist zone were more similar, while the distinction was increased with pollution levels among the tourist and mariculture zones. The 16S rRNA clone of D4 revealed that the Proteobacteria were the dominant phylum, and gamma-proteobacteria was the main class within Proteobacteria.

CONCLUSION

The study documented changes in bacterial community structure by human impacts of mariculture than geographical location.

目的

了解人为活动对细菌群落结构和组成的影响,并评估沿海沉积物中细菌群落如何响应环境梯度。

方法

采用末端限制性片段长度多态性(t-RFLP)和变性梯度凝胶电泳(DGGE)方法,评估大连长山群岛沿海旅游区和海水养殖区沉积物中细菌群落的多样性。同时,构建16S rRNA克隆文库,以揭示污染最严重的站点(D4)细菌群落的组成和结构。

结果

通过末端限制性片段(t-RFs)分析,D4站点的丰富度、香农-维纳指数和均匀度指数值更高。t-RFs面积和DGGE图谱的聚类结果表明,旅游区的细菌多样性更为相似,而旅游区和海水养殖区之间的差异随着污染程度的增加而增大。D4的16S rRNA克隆显示,变形菌门是优势菌门,γ-变形菌是变形菌门中的主要类群。

结论

该研究记录了海水养殖的人为影响比地理位置对细菌群落结构的改变更大。
