To evaluate the sensitization effect of different components of Shengmai injection (new production process) on Beagle dogs.
Beagle dogs were randomly divided into 7 groups, 3 in each group. Each group was respectively injected with 5% glucose injection, Ginseng Radix et Rhizoma Rubra extract, Ophiopogonis Radix extract, Schisandrae Chinensis Fructus extract, Schisandrae Chinersis Fructus distillate, Shengmaifang, 0.2% tween 80. The changes of each dog were observed from injection before until 24 hours after injection, and the response level was determined according to the severity of the symptoms. Blood samples were collected before injection and at 10 min after injection for measuring histamine content in plasma by ELISA. Sensitization of the injection was comprehensively determined by combined the response level of symptoms and the histamine level.
One dog of Ginseng Radix et Rhizoma Rubra extract group showed untypical symptoms of anaphylactoid reactions, and serum histamine of two dogs increased more than doubled. The Beagle dogs administrated with 0.2% tween 80 showed typical symptoms of anaphylactoid reactions, while there was no significant increase of serum histamine. Other groups were observed with no typical anaphylactoid reactions.
The sensitization effect of Shengmai injection (new production process) may be associated with Ginseng Radix et Rhizoma Rubra extract and 0.2% tween 80.
评价参麦注射液(新工艺)不同成分对比格犬的致敏作用。
将比格犬随机分为7组,每组3只。分别给每组犬注射5%葡萄糖注射液、红参提取物、麦冬提取物、五味子提取物、五味子馏出液、生脉方、0.2%吐温80。观察每只犬自注射前至注射后24小时的变化情况,并根据症状严重程度确定反应等级。于注射前及注射后10分钟采集血样,采用酶联免疫吸附测定法(ELISA)检测血浆组胺含量。结合症状反应等级和组胺水平综合判定注射液的致敏情况。
红参提取物组有1只犬出现非典型类过敏反应症状,2只犬血清组胺升高超过一倍。给予0.2%吐温80的比格犬出现典型类过敏反应症状,但血清组胺无明显升高。其他组未观察到典型类过敏反应。
参麦注射液(新工艺)的致敏作用可能与红参提取物和0.2%吐温80有关。
