Abstract

OBJECTIVES

To measure the levels of interleukin (IL)-27, interferon (IFN)-γ, and IL-17 in serum and supernatants of stimulated peripheral blood mononuclear cells (PBMCs) of patients with Behçet's disease (BD) compared with healthy subjects, and to evaluate the effect of recombinant human IL-27 (rhIL-27) on IFN-γ and IL-17 production by PBMCs.

METHODS

Forty-three patients with BD and 40 healthy controls were included in this study. The serum levels of IL-27, IFN-γ, and IL-17 and their production by PBMCs were measured by enzyme-linked immunosorbent assay (ELISA), together with the effects of levels of rhIL-27 on IFN-γ and IL-17 production by PBMCs.

RESULTS

There were significantly higher levels of IL-27, IFN-γ, and IL-17 in sera of patients with BD compared with the controls. Levels of IL-27, IFN-γ, and IL-17 in BD patients with active uveitis were significantly higher than in those without. Moreover, IL-27, IFN-γ, and IL-17 production by stimulated PBMCs was increased in BD patients compared to controls. In the presence of rhIL-27, stimulation of PBMCs from BD patients resulted in decreased production of IL-17 but increased production of IFN-γ.

CONCLUSIONS

These findings suggest that IL-27, IFN-γ, and IL-17 play a role in the pathogenesis of BD.

目的

比较 Behçet 病(BD)患者与健康对照者外周血单个核细胞(PBMC)刺激后血清和上清液中白细胞介素(IL)-27、干扰素(IFN)-γ和 IL-17 的水平,并评估重组人 IL-27(rhIL-27)对 PBMC 产生 IFN-γ和 IL-17 的影响。

方法

本研究纳入 43 例 BD 患者和 40 名健康对照者。采用酶联免疫吸附试验(ELISA)检测血清中 IL-27、IFN-γ和 IL-17 的水平及其 PBMC 的产生情况,并检测 rhIL-27 对 PBMC 产生 IFN-γ和 IL-17 的影响。

结果

BD 患者血清中 IL-27、IFN-γ和 IL-17 的水平明显高于对照组。活动期葡萄膜炎患者的 IL-27、IFN-γ和 IL-17 水平明显高于无葡萄膜炎患者。此外,与对照组相比,BD 患者刺激的 PBMC 产生的 IL-27、IFN-γ和 IL-17 增加。在 rhIL-27 存在的情况下,BD 患者 PBMC 的刺激导致 IL-17 的产生减少而 IFN-γ的产生增加。

结论
