Abstract

OBJECTIVE

To investigate the hypoglycemic effect of the aqueous extract of Octomeles sumatrana (O. sumatrana) (OS) in streptozotocin-induced diabetic rats (STZ) and its molecular mechanisms.

METHODS

Diabetes was induced by intraperitoneal (i.p.) injection of streptozotocin (55 mg/kg) in to male Sprague-Dawley rats. Rats were divided into six different groups; normal control rats were not induced with STZ and served as reference, STZ diabetic control rats were given normal saline. Three groups were treated with OS aqueous extract at 0.2, 0.3 and 0.5 g/kg, orally twice daily continuously for 21 d. The fifth group was treated with glibenclamide (6 mg/kg) in aqueous solution orally continuously for 21 d. After completion of the treatment period, biochemical parameters and expression levels of glucose transporter 2 (Slc2a2), glucose-6-phosphatase (G6Pase) and phosphoenolpyruvate carboxykinase (PCK1) were determined in liver by quantitative real time PCR.

RESULTS

Administration of OS at different doses to STZ induced diabetic rats, resulted in significant decrease (P<0.05) in blood glucose level in a dose dependent manner by 36%, 48%, and 64% at doses of 0.2, 0.3 and 0.5 g/kg, respectively, in comparison to the STZ control values. Treatment with OS elicited an increase in the expression level of Slc2a2 gene but reduced the expression of G6Pase and PCK1 genes. Morefore, OS treated rats, showed significantly lower levels of serum alanine transaminase (ALT), aspartate aminotransferase (AST) and urea levels compared to STZ untreated rats. The extract at different doses elicited signs of recovery in body weight gain when compared to STZ diabetic controls although food and water consumption were significantly lower in treated groups compared to STZ diabetic control group.

CONCLUSIONS

O. sumatrana aqueous extract is beneficial for improvement of hyperglycemia by increasing gene expression of liver Slc2a2 and reducing expression of G6Pase and PCK1 genes in streptozotocin-induced diabetic rats.

方法

雄性 Sprague-Dawley 大鼠腹腔注射链脲佐菌素(55mg/kg)诱导糖尿病。大鼠分为六组;正常对照组大鼠不给予 STZ 作为参考,STZ 糖尿病对照组给予生理盐水。三组以 0.2、0.3 和 0.5g/kg 的剂量口服 OS 水提物,每天两次,连续 21d。第五组给予口服水溶液中的格列本脲(6mg/kg),连续 21d。治疗期结束后,通过定量实时 PCR 测定肝中葡萄糖转运蛋白 2(Slc2a2)、葡萄糖-6-磷酸酶(G6Pase)和磷酸烯醇丙酮酸羧激酶(PCK1)的生化参数和表达水平。

结果

不同剂量 OS 给药至 STZ 诱导的糖尿病大鼠,以剂量依赖性方式使血糖水平显著降低(P<0.05),0.2、0.3 和 0.5g/kg 剂量分别降低 36%、48%和 64%,与 STZ 对照组相比。OS 处理诱导 Slc2a2 基因的表达水平增加,但降低 G6Pase 和 PCK1 基因的表达。此外,OS 处理的大鼠与未处理 STZ 的大鼠相比,血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)和尿素水平显著降低。与 STZ 糖尿病对照组相比,不同剂量的提取物在体重增加方面表现出明显的恢复迹象,尽管与 STZ 糖尿病对照组相比,处理组的食物和水消耗明显减少。
