To develop a high performance liquid chromatography (LC) method for simultaneously determination of polycyclic aromatic hydrocarbons (PAHs) in serum.
PAHs could be separated completely by using C18 column and detected by fluorescence detector at wavelengths (Ex = 340 nm, Em = 425 nm). The mobile phase A was acetonitrile and mobile phase B was water with a flow rate at 1.0 ml/min.
The concentration of PAHs achieved a good linear relation with the peak area. The correlation coefficient was in the range of 0.9941-0.9999. The recoveries were in the range of 91.4%-99.8% and the RSD were in the range of 2.56%-12.7%. The detection limits were in the range of 0.05-0.10 ng/ml.
The method was simple, rapid, sensitive and accurate, and suitable for the detection of PAHs in serum.
建立一种同时测定血清中多环芳烃(PAHs)的高效液相色谱(LC)方法。
采用C18柱可将PAHs完全分离,并用荧光检测器在波长(激发波长Ex = 340 nm,发射波长Em = 425 nm)下进行检测。流动相A为乙腈,流动相B为水,流速为1.0 ml/min。
PAHs浓度与峰面积呈良好的线性关系。相关系数在0.9941 - 0.9999范围内。回收率在91.4% - 99.8%范围内,相对标准偏差(RSD)在2.56% - 12.7%范围内。检测限在0.05 - 0.10 ng/ml范围内。
该方法简便、快速、灵敏、准确,适用于血清中PAHs的检测。
