The growth-limiting reaction of a chemostat culture of Escherichia coli K-12 was the hydrolysis of beta-glycerophosphate by alkaline phosphatase. The culture was buffered at pH 5.2 where alkaline phosphatase was unable to supply phosphate to the cell at a rate sufficient to sustain the maximum rate of growth. Alkaline phosphatase activity in this system is discussed in terms of the so-called Flip-Flop mechanism.
