Abstract

OBJECTIVES

Head and neck squamous cell carcinomas (HNSCCs) are characterized by marked heterogeneity in their biological behavior and response to treatment. Our goal was the identification of biomarkers that can be used to predict response to chemotherapy in these patients.

MATERIALS AND METHODS

The expression of EGFR, p53, Cyclin D1, p16, p21, p27, p-AKT, HIF-1α, Caspase 3 and BCL2 was analyzed by immunohistochemistry in 41 primary laryngeal/hypopharyngeal squamous cell carcinomas of patients that received induction chemotherapy (cisplatin and 5-fluorouracil) as part of their treatment.

RESULTS

Positive expression of p27 and BCL2 had a significant predictive value for chemotherapy response in univariate analysis. The combination of both proteins was not superior in predicting the response to chemotherapy. Furthermore, p27 expression was the only significant predictor of chemotherapy response in multivariate analysis (P=0.015).

CONCLUSION

p27 Expression may serve as predictive biomarker of response to induction chemotherapy in HNSCC patients.

目的

头颈部鳞状细胞癌(HNSCCs)的生物学行为和对治疗的反应具有显著异质性。我们的目标是鉴定可用于预测这些患者化疗反应的生物标志物。

材料与方法

通过免疫组织化学分析了41例接受诱导化疗(顺铂和5-氟尿嘧啶)作为治疗一部分的原发性喉/下咽鳞状细胞癌患者中EGFR、p53、细胞周期蛋白D1、p16、p21、p27、p-AKT、HIF-1α、半胱天冬酶3和BCL2的表达。

结果

在单因素分析中,p27和BCL2的阳性表达对化疗反应具有显著预测价值。两种蛋白的联合在预测化疗反应方面并不更具优势。此外,在多因素分析中,p27表达是化疗反应的唯一显著预测因子(P = 0.015)。

结论

p27表达可能作为HNSCC患者诱导化疗反应的预测生物标志物。
