Sirtuin 1 (SIRT1) has been reported to have diverse roles in various biological processes through deacetylation of histone and nonhistone proteins. However, the correlations among SIRT1 protein expression, clinicopathological parameters, and survival of colorectal cancer patients remain unclear.
SIRT1 protein expression was measured by immunohistochemistry in a paraffin-embedded tissue microarray, including 120 paired colorectal cancer and normal mucosa tissues. The correlations among SIRT1 protein expression, clinicopathological features, and prognosis were analyzed.
All samples (100%) were positive for SIRT1, with variable staining in the cytoplasm rather than in the nucleus. There was significant difference in SIRT1 overexpression between adenocarcinomas and normal mucosal tissue (P < 0.01, χ(2) test). SIRT1 overexpression was more frequently observed in advanced-stage tumors (P = 0.046, 0.002, χ(2) test). SIRT1 overexpression was significantly correlated with poor overall survival (P = 0.013, log-rank test) and diseasefree survival (P = 0.012, log-rank test).
SIRT1 overexpression correlated with advanced stage and poor prognosis. SIRT1 may play an important role in the progression of colorectal cancer.
据报道,沉默调节蛋白1(SIRT1)通过对组蛋白和非组蛋白进行去乙酰化作用,在多种生物学过程中发挥不同作用。然而,SIRT1蛋白表达、临床病理参数与结直肠癌患者生存率之间的相关性仍不明确。
采用免疫组织化学法检测石蜡包埋组织芯片中120对结直肠癌组织和正常黏膜组织中SIRT1蛋白的表达。分析SIRT1蛋白表达、临床病理特征与预后之间的相关性。
所有样本(100%)SIRT1均呈阳性,染色位于细胞质而非细胞核,且存在差异。腺癌与正常黏膜组织之间SIRT1过表达存在显著差异(P < 0.01,χ²检验)。在晚期肿瘤中更常观察到SIRT1过表达(P = 0.046,0.002,χ²检验)。SIRT1过表达与总生存期差(P = 0.013,对数秩检验)和无病生存期差(P = 0.012,对数秩检验)显著相关。
SIRT1过表达与晚期和预后不良相关。SIRT1可能在结直肠癌进展中起重要作用。
