Abstract

OBJECTIVE

To investigate the effect of curcumin (Cur) on radiosensitivity of nasopharyngeal carcinoma cell CNE-2 and its mechanism.

METHOD

The effect of curcumin on radiosensitivity was determined by the clone formation assay. The cell survival curve was fitted by Graph prism 6. 0. The changes in cell cycle were analyzed by flow cytometry (FCM). The differential expression of long non-coding RNA was detected by gene chip technology. Part of differentially expressed genes was verified by Real-time PCR.

RESULT

After 10 micro mol L-1 Cur had worked for 24 h, its sensitization enhancement ratio was 1. 03, indicating that low concentration of curcumin could increase the radiosensitivity of nasopharyngeal carcinoma cells; FCM displayed a significant increase of G2 phase cells and significant decrease of S phase cells in the Cur combined radiation group. In the Cur group, the GUCY2GP, H2BFXP, LINC00623 IncRNA were significantly up-regulated and ZRANB2-AS2 LOC100506835, FLJ36000 IncRNA were significantly down-regulated.

CONCLUSION

Cur has radiosensitizing effect on human nasopharyngeal carcinoma CNE-2 cells. Its mechanism may be related to the changes in the cell cycle distribution and the expression of long non-coding IncRNA.

目的

探讨姜黄素(Cur)对鼻咽癌CNE-2细胞放射敏感性的影响及其机制。

方法

采用克隆形成实验检测姜黄素对放射敏感性的影响。用Graph prism 6.0拟合细胞存活曲线。采用流式细胞术(FCM)分析细胞周期变化。利用基因芯片技术检测长链非编码RNA的差异表达。部分差异表达基因通过实时荧光定量PCR进行验证。

结果

10 μmol/L Cur作用24 h后,其增敏增强比为1.03,表明低浓度姜黄素可提高鼻咽癌细胞的放射敏感性;FCM显示Cur联合放疗组G2期细胞显著增多,S期细胞显著减少。在Cur组中,GUCY2GP、H2BFXP、LINC00623 IncRNA显著上调,ZRANB2-AS2 LOC100506835、FLJ36000 IncRNA显著下调。

结论

Cur对人鼻咽癌CNE-2细胞有放射增敏作用。其机制可能与细胞周期分布改变及长链非编码IncRNA表达有关。
